The Automated GX-271 GPC Clean-up System provides automated post-extraction clean-up of interfering substances from environmental samples, food products and animal tissue by Gel Permeation Chromatography (GPC) prior to analysis by GC, GC/MS, HPLC or LC/MS. Samples that are extracted from foods, animal and plant tissue, soils, sediment and wastewater contain a number of substances (such as lipids, proteins, cell debris, pigments, humic acid, etc.) that can interfere with the analysis for pharmaceuticals and residues such as pesticides, PCBs, antibiotics, mycotoxins, polyaromatic hydrocarbons (PAHs) and other semi-volatile compounds. Failure to remove these interfering compounds can cause poor analytical results, shorten column life and increase analytical downtime. The Gilson Automated GX-271 GPC Clean-up System is ideal for the automated removal of interfering substances. The system is controlled with TRILUTION® LC Software, a user-friendly, easy-to-use, flexible program that was specifically designed to meet the laboratory needs. The software features pre-programmed methods specific to GPC post-extraction clean-up processes.

Concept

GPC is a size exclusion clean-up procedure that uses organic solvents for the mobile phase and a specialized column packed with a divinylbenzene-styrene copolymer. The most typical packed phase uses Bio Beads SX-3 resin, packed into a glass column with the mobile phase of interest. Stainless steel and other columns specific for GPC clean-up are also available. GPC clean-up is performed after sample extraction, and is thus a post-extraction clean-up method. This method is recommended for the post-extraction elimination of lipids, polymers, copolymers, proteins, natural resins, cellular components and other high molecular weight compounds from the analytes of interest.

The column is typically flushed with an organic solvent such as dichloromethane and then the column is calibrated. Calibration is usually performed once per week. Sample extracts are then transferred into vials (extracts are prepared in the GPC mobile phase) and then placed on the GX-271 Liquid Handler rack. The Direct Injection Module introduces the extracts into the column. The mobile phase is always isocratic. High
molecular weight materials, such as lipids, elute first and are discarded to waste. The remaining sample with the analytes of interest is collected into a large vessel on a collection tray for further processing (50-200 mL are the typical collection volumes). Late-eluting fractions, such as sulfur in soil/sediment extracts, can be directed to waste after collecting the compounds of interest. The system is completely rinsed between sample injections.

Product Design

The Automated GX-271 GPC Clean-up System is available in two configurations, with a UV Detector and without. The system configured with the 112 UV Detector with Prep Flow Cell includes a GX-271 Liquid Handler without pump, GX Direct Injection Module, GX-271 GPC Clean-up Accessory Kit, 402 Syringe Pump (single module), 307 Pump Module, 506C System Interface Module, Autozero and detector cables, TRILUTION® LC Media v1.4 with license and ORACLE Database for GPC Control as well as a CD that includes GPC methods and instructions.

The UV detector can be utilized by laboratories complying with GPC post-extraction guidelines set by the U.S. Environmental Protection Agency. The UV detector is used to calibrate the GPC column once a week ensuring regulation standard (see Control Software page 11). The Automated GX-271 GPC Clean-up System without UV detector includes a GX-271 Liquid Handler, GX Direct Injection Module, GX-271 GPC Clean-up Accessory Kit, 402 Syringe Pump (single module), 307 Pump Module, 508 System Interface Module,TRILUTION® LC Media v1.4 with license and ORACLE Database for GPC Control as well as a CD that includes GPC example methods and instructions. This system would be most commonly used for methods written for post-extraction clean-up of food matrices. These two configurations fully comply with USEPA, USFDA, USDA, USGS, ISO, CFIA, AOAC International and European Union (EN 1528, EN 12393, S19) and other government agency guidelines for GPC post-extraction clean-up.

GPC Control Software

The Automated GX-271 GPC Clean-up System is controlled via TRILUTION® LC Software. Methods specifically designed to meet the needs of GPC post-extraction clean-up are programmed in the software for ease of use. A CD-ROM including instructions for proper system installation, a TRILUTION® LC Software database file that contains GPC clean-up example methods for the Automated GX-271 GPC Clean-up System and software instructions are included with each order. There are separate methods supplied for GPC systems with the 112 UV detector.
